Volume enhancement

The low SNR and the presence of irrelevant but intensity-similar structures near the microtubules necessitate a preprocessing step to enhance the microtubules before segmentation. We developed a hybrid method to exploit the geometric properties of the tubular objects by combining a transform domain technique and spatial domain techniques:

 

  • Anisotropic invariant wavelet transform

-- enhance elongated objects globally, attenuate irrelevant features 

  • 3D Tube-enhancing filter

-- enhance microtubules locally from low contrast image

  • Modified coherence-enhancing diffusion

-- complete interruptions along the microtubules

  • 3D Surface-enhancing filter

    enhance fine surface features in localized volume
